چکیده
The Doxorubicin hydrochloride, popularly known as Adriamycin, is among the most antitumoral drugs used in oncology practice. This is a glycosidic antibiotic from the anthracycline group, which acts directly on the G2/M period, leading to cell death. This study aimed at analyzing placental alteration and congenital malformations in Wistar rats fetuses who were exposed to the chemotherapeutic. Twelve Wistar rats were divided into two groups: Control (n = 6) and Treated (n = 6) and were exposed to Adriamycin on the 8th and 9th days of pregnancy. Both morphological and morphometric analyses in the placenta were performed, the fetuses were weighed, measured and macroscopically assessed for the presence of congenital malformations, which, once occurring were photographed. The results indicate intense thickness of the spongy zone (p < 0.05), disarrangement of the labyrinth with blood extravasation from the fetal vessels, besides the presence of infiltrated leukocytic. Fetuses weight and length in the treated group were lower than in the control group (p < 0.01). The congenital malformations such as edema, ear low implantation, twisted tail, short tail, dysmorphic eyes, occurred in 23 (82%) out of the 28 embryos in the Adriamycin group, against 14 (19%) out of 72 embryos in the control group Thus, we can infer that Adriamycin increases the incidence of congenital malformations, mainly those related to the bone tissue. In addition, it provokes embryo reabsorption in some cases, impairs the process of maternal-fetal exchange in the placenta and contributes to the restriction of intrauterine growth.

Two zebrafish Notch-dependent hairy/Enhancer-of-split-related genes, her6 and her4, are required to maintain the coordination of cyclic gene expression in the presomitic mesoderm.
Alterations of the Delta/Notch signalling pathway cause multiple morphogenetic abnormalities in somitogenesis, including defects in intersomitic boundary formation and failure in maintenance of somite regularity. Notch signalling has been implicated in establishing the anteroposterior polarity within maturing somites and in regulating the activity of a molecular segmentation clock operating in ...
متن کاملtbx2a Is Required for Specification of Endodermal Pouches during Development of the Pharyngeal Arches
Tbx2 is a member of the T-box family of transcription factors essential for embryo- and organogenesis. A deficiency in the zebrafish paralogue tbx2a causes abnormalities of the pharyngeal arches in a p53-independent manner. The pharyngeal arches are formed by derivatives of all three embryonic germ layers: endodermal pouches, mesenchymal condensations and neural crest cells. While tbx2a express...
متن کاملHistone deacetylase 1 is required to repress Notch target gene expression during zebrafish neurogenesis and to maintain the production of motoneurones in response to hedgehog signalling.
Histone deacetylases (Hdacs) are widely implicated as key components of transcriptional silencing mechanisms. Here, I show that hdac1 is specifically required in the zebrafish embryonic CNS to maintain neurogenesis. In hdac1 mutant embryos, the Notch-responsive E(spl)-related neurogenic gene her6 is ectopically expressed at distinct sites within the developing CNS and proneural gene expression ...
متن کاملTBX1, a DiGeorge syndrome candidate gene, is inhibited by retinoic acid.
Both retinoic acid (RA) and Tbx1 are definitively indispensable for the development of the pharyngeal arches. The defects produced by a loss of Tbx1 highly resemble those induced by hyper- and hypo-RA. Based on these similarities, the effects of RA on Tbx1 expression pattern were explored during pharyngeal arch development in zebrafish. Whole-mount in situ hybridization and real-time quantitati...
متن کاملThe zebrafish van gogh mutation disrupts tbx1, which is involved in the DiGeorge deletion syndrome in humans.
The van gogh (vgo) mutant in zebrafish is characterized by defects in the ear, pharyngeal arches and associated structures such as the thymus. We show that vgo is caused by a mutation in tbx1, a member of the large family of T-box genes. tbx1 has been recently suggested to be a major contributor to the cardiovascular defects in DiGeorge deletion syndrome (DGS) in humans, a syndrome in which sev...
